<p class="wp-block-paragraph">Right now, one of the biggest and most controversial topics in biomedical research is Artificial Intelligence. Modern AI systems, based on Machine Learning (ML) models, are being increasingly used in speculative research.</p>



<p class="wp-block-paragraph">This has led to some genuinely interesting discoveries, such as discovering more <a href="https://www.nature.com/articles/s41588-019-0420-0">genetic basis</a> for autism spectrum conditions. AI in medical device development shows similar&nbsp;promise, such as the development of an <a href="https://www.cardiocaretoday.com/post/researchers-use-ai-and-wrist-worn-sensor-to-detect-heart-disease">AI-powered wristband</a>&nbsp;capable of monitoring for heart disease. AI is also being explored for its potential to speed up the processing of data and the creation of publication-ready research materials.</p>



<p class="wp-block-paragraph">However, there are still numerous issues with AI in biomedical research, which are perhaps being overlooked in the current &#8216;gold rush&#8217; environment. More critically, there are hard limits in what AI can accomplish. There is a barrier between theoretical AI models and practical real-world testing, which will likely never go away.</p>



<p class="wp-block-paragraph">To be clear, we are not anti-AI; any research that improves human medicine is welcome. However, we know that the potential drawbacks and challenges facing AI medical research must be discussed. So, in this article, we&#8217;ll look at some of the biggest issues with current AI research and why real tissue testing using actual animal materials remains vital.</p>



<h2 class="wp-block-heading"><em>In Silico&nbsp;</em>vs&nbsp;<em>In Vitro&nbsp;</em>Research &#8211; Can a&nbsp;Computer Model Cellular Activity?</h2>



<p class="wp-block-paragraph">It&#8217;s easy to understand the appeal of computer models. If a computer could accurately simulate a complicated biological process, sometimes called &#8220;<em>in silico</em>&#8221; research, then research could be accelerated. Better yet, it would require less use of animal or human test subjects, making it a theoretically superior ethical choice.</p>



<p class="wp-block-paragraph">Unfortunately, there are limits to how far this could go. Due to the complexity of cells and biological processes, computational biology has its limits. A <a href="https://www.pnas.org/doi/10.1073/pnas.2022753118">2021 PNAS article&nbsp;</a>estimated that it would be decades, if ever, before computers were capable of fully simulating multicellular systems.</p>



<p class="wp-block-paragraph">Further, that assumes biological processes operate according to a standard model. While currently&nbsp;under debate, recent research suggests that&nbsp;<a href="https://www.eurekalert.org/news-releases/1077836">quantum effects</a> may be involved in biological processes. If true, that would imply that perfect&nbsp;<em>in silico&nbsp;</em>models may be practically impossible, even with quantum computing.</p>



<p class="wp-block-paragraph">In short, while AI has proven effective at analyzing large existing data sets, its&nbsp;<em>predictive&nbsp;</em>abilities may be severely limited.</p>

<h2 class="wp-block-heading">The Black Box Problem &#8211; What&#8217;s That AI Doing, Anyway?</h2>

<p class="wp-block-paragraph">Another major issue exists, as discussed in <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC9908503/">Biomaterial Medical Devices in 2023</a> &#8211; the &#8220;black box&#8221; nature of AI reasoning. Simply put, the way current Machine Learning&nbsp;models work is extremely fuzzy. It is often difficult or impossible to understand exactly why or how&nbsp;an AI model came to a certain conclusion.</p>



<p class="wp-block-paragraph">Unlike a grad student or junior researcher, you cannot ask an AI to explain its &#8216;thought process&#8217; step by step to analyze its reasoning. Current AI systems based on Large Language Models (LLMs) are&nbsp;incapable of that sort of self-reflection.</p>



<p class="wp-block-paragraph">Worse, in some cases, it&#8217;s possible to feed the same data into the same AI and have it reach two different conclusions. Again, with no reasoning to analyze.</p>



<p class="wp-block-paragraph">This could have a significant impact on the ability of researchers to turn AI insights into real-world research utilizing real tissue testing, much less live subjects. No translational research tools exist that can turn a &#8216;black box&#8217; discovery into a research proposal that appears to have a firm basis. At best the research would be purely exploratory, effectively saying, &#8220;The AI thinks this is a good idea, so let&#8217;s give it a shot.&#8221;</p>



<p class="wp-block-paragraph">Such proposals may be viable with basic tissue-based research methods involving post-mortem materials, but would still face heightened scrutiny. If a researcher is unable to explain how a proposed outcome might occur, research will be slowed.</p>



<h2 class="wp-block-heading">How Can Potential Biases of AI in Biomedical Research Countered?</h2>



<p class="wp-block-paragraph">Another important issue raised by the above&nbsp;<a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC9908503/">Biomed Mater Devices article</a>&nbsp;is the problem of human biases slipping into AI research. We like to think of computers as being impartial, and they are in some ways.</p>



<p class="wp-block-paragraph">However, an AI is only as unbiased as its data set. Online retailer <a href="https://www.reuters.com/article/world/insight-amazon-scraps-secret-ai-recruiting-tool-that-showed-bias-against-women-idUSKCN1MK0AG/">Amazon discovered this the hard way a few years ago</a>, when a system set up to analyze resumes in hiring began discriminating against women. Because many resumes fed to the system were from men, the system took this to mean that male candidates&nbsp;were preferable.</p>



<p class="wp-block-paragraph">Likewise, medical data can skew&nbsp;along racial lines. Black women have a 38% higher fatality rate from breast cancer than white women, yet they only represent <a href="https://www.bcrf.org/about-breast-cancer/breast-cancer-racial-disparities/">five percent</a>&nbsp;of research subjects. This undoubtedly influences research outcomes.</p>



<p class="wp-block-paragraph">An ML AI may be completely incapable of recognizing these bias issues. Thus, without rigorous data scrubbing and validation far beyond current standards, human biases will still creep into AI analyses.</p>



<h2 class="wp-block-heading">Real Tissue Testing Will Always Be Needed for Translational Research</h2>

While this may seem counterintuitive, there are very good reasons for it. Hands-on training is a must for surgical training, but there are numerous challenges in obtaining and utilizing human remains. On the other hand,  <a href="https://animalbiotech.com/post-mortem-porcine-tissues/">post-mortem swine tissues</a> are widely seen as the best alternative to training on human remains, and with a number of other benefits as well.</p>



<p class="wp-block-paragraph">In this article, we&#8217;ll be looking at the effectiveness of&nbsp;porcine organs used for surgical training and skills labs, and how they can benefit training operations.</p>

<li><strong>Spinal surgery training: </strong>Minimally invasive surgery training for spinal work previously lacked good alternatives to human cadavers, but recent <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC6857820/">studies</a> have shown positive results from swine-based training as well.</li>

<p class="wp-block-paragraph">The extracellular matrix (ECM) is a complicated system of proteins and carbohydrates that are excreted by cells as a support system. The ECM aids in&nbsp;a wide variety of functions, including cell communication, movement, and &#8211; most relevant here &#8211; repair.</p>



<p class="wp-block-paragraph">When tissue takes damage, the ECM could be seen as constructing a sort of scaffold that guides tissue repair. The problem is, the more traumatic the damage, the more difficult this process becomes. This&nbsp;slows&nbsp;healing and increases the chances of comorbid factors such as infection.</p>



<p class="wp-block-paragraph">So, finding ways to support and speed up the healing of such wounds is of great interest to researchers.&nbsp;However, the matter is complicated in several ways.</p>



<p class="wp-block-paragraph">Each cell or tissue type tends to have a unique ECM composition, and the ECM directly influences cell growth/regrowth. So, any attempt to modify or replace the ECM cannot impede or sabotage cell regrowth. Further, there are immunological considerations. Any outside aids must be accepted by the body, as immune rejection would only cause more damage.</p>



<p class="wp-block-paragraph">In recent years, study has turned towards the use of decellularized&nbsp;donor tissue, such as post-mortem <a href="https://animalbiotech.com/2025/05/27/revolutionizing-lung-tissue-repair-the-potential-of-decellularized-porcine-pleura/">porcine lung tissue</a>, as a source of donor ECM.&nbsp;Decellularized ECM (dECM) has great potential to aid in injury recovery, especially if methods can be found to prevent rejection while properly aiding tissue regrowth.</p>



<h2 class="wp-block-heading">II. How is Decellularized ECM Created?</h2>



<p class="wp-block-paragraph">The decellularization process involves stripping the donor tissue of its&nbsp;immunogenic cellular components &#8211; such as DNA &#8211; which can trigger immune responses in recipients, while still preserving the underlying ECM structure.</p>



<p class="wp-block-paragraph">Per the <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC12565388/">Journal of Functional Biomaterials</a>&nbsp;in 2025, the current most common way of achieving this is through &#8220;ionic detergents such as sodium dodecyl sulfate (SDS), sodium deoxycholate (SDC), and Triton X-100,&#8221; which are highly effective at removing cellular components.&nbsp;However, the article notes that&nbsp;the detergents&nbsp;must be precisely applied to avoid binding with the remaining cells and causing cytotoxic effects.</p>



<p class="wp-block-paragraph">Still,&nbsp;ionic detergents&nbsp;are seen as preferable to less aggressive methods that often fail to properly scrub the tissue of genetic material, requiring multiple processes to complete the decellularization.&nbsp;</p>



<p class="wp-block-paragraph">In particular, this ionic detergent process is cited as highly successful on post-mortem porcine tissue in a 2025 Biomaterial Advances <a href="https://www.sciencedirect.com/science/article/pii/S2772950825002092">article</a>. In the documented experiment, they were able to create dECM material suitable for human cardiovascular repair from porcine&nbsp;liver.&nbsp;</p>



<p class="wp-block-paragraph">This is one of the reasons swine are now becoming a common source for biomedical scaffolds.</p>

<p class="wp-block-paragraph">In general, swine have been noted for having a biology remarkably similar to humans, even more so than species one would expect to be a close match, such as great apes. From genetic components to basic organ size and function, swine organs and tissues are already a close match to humans.</p>



<p class="wp-block-paragraph">This has made swine one of the most popular sources for human medical research, in applications ranging&nbsp;from <a href="https://animalbiotech.com/2021/04/20/xenotransplantation-the-future-of-skin-grafts/">skin grafts</a>&nbsp;to <a href="https://animalbiotech.com/2025/01/20/pigskin-corneal-implants-a-new-frontier-in-ophthalmology/">corneal replacement</a>.</p>



<p class="wp-block-paragraph">The same is holding true for the use of porcine-derived tissue engineering scaffolds and other dECM applications. In 2022, a Modern Technologies in Medicine <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC10090917/">article</a>&nbsp;cited multiple&nbsp;successful uses of porcine-derived dECM including replacing joint cartilage, as well as noting it was currently the most popular source of dECM materials.</p>



<p class="wp-block-paragraph">At least one <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC3950205/">study</a> &#8211; published in&nbsp;Biomaterials Science &#8211; even found that porcine materials produced superior results to human-derived alternatives for injectable biomaterial.</p>



<p class="wp-block-paragraph">Synthetic materials are&nbsp;another alternative for creating ECM scaffolds, but come with their own issues.&nbsp;They&nbsp;struggle due to their lack of bioactivity and a tendency to provoke immune responses.&nbsp;As described&nbsp;in a separate J Funct Biomater <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC12653107/">article</a>, &#8220;porcine-derived scaffolds strike a balance between biologic performance and clinical manageability.&#8221; However, the article&nbsp;does note that synthetic ECM&nbsp;scaffolds do have some advantages in strength and flexibility.</p>

<p class="wp-block-paragraph">There&#8217;s also the matter of availability. Human-derived donor material is always difficult to come by, and demand frequently outpaces supply,&nbsp;often by a high degree. In all likelihood, there will always be fewer donors than people in need of donated human tissue.</p>



<p class="wp-block-paragraph">Meanwhile, swine are among the most farmed, bred, and researched animals on Earth. <p class="wp-block-paragraph">Today, more than <a href="https://pmc.ncbi.nlm.nih.gov/articles/PMC5820628/figure/F4/">250 million people</a> worldwide are blind or live with significant vision loss that cannot be corrected with lenses. With longer life expectancies and limited access to specialized eye care, the demand for new vision treatments is higher than ever, and researchers need consistent access to quality biological material to make progress.</p>



<h2 class="wp-block-heading">Why Is Human Eye Tissue Hard to Source for Research?</h2>



<p class="wp-block-paragraph">A <a href="https://www.nature.com/articles/s41598-025-92291-3">2025 study in </a><a href="https://www.nature.com/articles/s41598-025-92291-3">Nature</a>&nbsp;revealed the extent of the problems faced by researchers who need access to post-mortem human eyes. Because the eye breaks down so quickly after death, retrieval of donor tissue must be done rapidly&nbsp;<em>and&nbsp;</em>transported rapidly as well, to be useful to researchers.</p>



<p class="wp-block-paragraph">However, based on recoveries at a U.S. eye bank across 2022–23, these goals are rarely met:</p>



<ul class="wp-block-list">
<li>Only <a href="https://www.nature.com/articles/s41598-025-92291-3#:~:text=Among%20the%20eyes%20recovered%20specifically%20for%20research%2C%20only%2032%25%20of%20tissues%20met%20the%20%3C%E2%80%8912%C2%A0h%20PMI%20requirement%20commonly%20requested%20by%20researchers.">32% of donor eyes were recovered</a> and shipped within the preferred 12-hour post-death timeframe.</li>



<li>Even fewer (just 27%) were successfully delivered during standard working hours when trained technicians were available.</li>
</ul>



<p class="wp-block-paragraph">So even if a donor has allowed their eyes to be used by science, well under one-third are actually utilized.</p>



<p class="wp-block-paragraph">The research points to specific policy changes that could improve these numbers by increasing office hours as well as being more lenient about post-death shipping times. However, even then, a substantial number of eyes would still go unused.</p>



<p class="wp-block-paragraph">This would logically result in a reduction in research, an assumption backed up by a <a href="https://pubmed.ncbi.nlm.nih.gov/30304462/">2018&nbsp;survey</a> of&nbsp;researchers attempting to source human ocular&nbsp;tissue. 43% reported that it was &#8220;difficult&#8221; or &#8220;very difficult&#8221; to obtain human eyes. Similar numbers also reported that they had to scale back research due to tissue unavailability or &#8211; perhaps worse &#8211; that they doubted the quality of the tissue they were working with.&nbsp;</p>



<p class="wp-block-paragraph">In short, the difficulties of vision science tissue sourcing&nbsp;have&nbsp;become a significant barrier to quality research, even as the need for such research continues to increase globally.</p>



<h2 class="wp-block-heading">What Are the Best Alternatives to Human Eye Tissue?</h2>



<p class="wp-block-paragraph">Fortunately, there is a viable solution: <a href="https://animalbiotech.com/porcine-tissues/sensory-organs/">porcine eyes for research</a>.</p>

<h2 class="wp-block-heading">A Bioscience Career Story: Operations Manager Brendan Lederach</h2>



<p class="wp-block-paragraph">As Brendan tells it, his love of animals is truly lifelong &#8211; ever since he was three years old and rescued a stray kitten near his town&#8217;s community pool.</p>



<p class="wp-block-paragraph">Growing up in the country, he was able to keep that connection with animals going. In school, he was part of the <a href="https://4-h.org/">4H Club</a>, raising chickens and rabbits for projects while gaining an appreciation for the ethical treatment of farmed animals.</p>



<p class="wp-block-paragraph">In addition, he was an <a href="https://nesa.org/">Eagle Scout</a>&nbsp;and enjoyed numerous trips into the countryside while picking up merit badges. Along the way, he became interested in hunting &#8211; not merely for sport but cultivating an understanding of the circle of life. Even from a young age, he was focused on ensuring that every animal killed was put to the best use possible, so that their sacrifice could help others.</p>



<p class="wp-block-paragraph">However, his true turning point was when he began raising seeing-eye dogs alongside his parents. That was when he realized that caring for animals could truly change people&#8217;s lives for the better. This realization would push him forward in the years to come.&nbsp;</p>



<p class="wp-block-paragraph">From there, he went to college at <a href="https://delval.edu/">Delaware Valley University</a>. Originally, he was thinking of becoming a vet, but discovered he had more interest in the business side of agriculture. So, he majored in Agribusiness&nbsp;before discovering ABI during his senior year of college.</p>



<p class="wp-block-paragraph">Brendan&nbsp;graduated in 2014 and was already working here the next spring. Our passion for ethical tissue sourcing for the sake of better bioresearch spoke to him, meshing perfectly with his own love of human/animal partnerships,&nbsp;and he found his professional home.</p>
